Abstract Gene coding genes have a high amino acid sequence homology and are expressed under the control of promoter and enhancer sequences but they usually are expressed by directly sponging RNA molecules called silencing complexes, a phenomenon in which RNA molecules are transferred by plasmid-mediated RNA pulses to the nucleus in response to DNA polymerase, which is synthesized in the nucleus and subsequently degraded into RNA strands. While silencing is not crucial for the establishment or maintenance of gene expression, it has been recognized that in contrast to silenced genes they could play a role in the growth, proliferation and differentiation of cells as well as the recovery of tissues after tumorigenesis. To test this idea we have studied the effect of two genes, *Rb1* and *K5*, on the recovery, organization and transcriptional program of their mRNA transcripts. Results *Rb1* does not show any changes in expression relative to *Fgf2* but the fact that the *K5* mRNA transcript is very poorly expressed reflects this. GFP transcripts can be recovered from the nucleus of live cells prior to nuclear transfer but endogenous expression of *Rb1* in this nucleus is not changed from the time of nuclear transfer. Therefore *Rb1* expression in the nucleus of HEK293T cells is minimal (at 2% relative to untreated controls) and *K5* is the only expressed gene that is completely transcriptionally active. A possible reason for this observation is that *K5* affects transcription of its own mRNA but the majority of the genes that produce the transcript are *Rb1* transcripts.
